摘要
目的观察5,7,4’-三羟基异黄酮(Genistein)对体外培养的人瘢痕疙瘩成纤维细胞生长及侵袭作用的影响,探讨其抗纤维化作用的机制。方法以25、50、100μmol/L浓度Genistein处理体外培养的人瘢痕疙瘩成纤维细胞,MTT法检测细胞增殖活性,3H-脯氨酸掺入法检测细胞胶原合成,Transwell小室趋化运动模型检测细胞侵袭能力。结果Genistein作用后,瘢痕疙瘩成纤维细胞的增殖及胶原合成作用降低,细胞的侵袭能力显著下降。随着药物浓度增加,对细胞的这种抑制作用也增强。结论Genistein具有体外抗瘢痕疙瘩成纤维细胞纤维化与侵袭的作用,可成为治疗病理性瘢痕及纤维化疾病的有效药物。
Objective To observe the effects of genistein on the proliferation and invasion of the cultured human keloid fibroblasts and explore the mechanism of its anti-fibrosis. Methods The human keloid fibroblasts were cultured and stimulated with 25, 50, 100 moL/L genistein, respectively. Their proliferation was measured by MTT. The collagen synthesis was assessed by ^3H-proline incorporation method. The invasion of human keloid fibroblasts was detected in Transwell chemotaxis chamber model. Results Genistein markedly inhibited the proliferation and collagen synthesis of the cultured keloid fibroblasts and the inhibitory effects were increased with the increasing concentration of genistein. Genistein also weakened the invasion of keloid fibroblasts. Conclusion Genistein has the ability of anti-fibrosis and inhibits the invasion of human keloid fibroblasts in vitro, indicating genistein may be an effective drug for pathological scar or fibrosis disease.
