非小细胞肺癌患者外周血中CK19 mRNA、MUC-1 mRNA及LUNX mRNA的表达及意义 被引量:4

摘要 目的应用巢式逆转录聚合酶链反应(Nested RT-PCR)技术检测非小细胞肺癌患者外周血中CK19mRNA、MUC-1mRNA及LUNXmRNA的表达,探讨其作为肺癌微转移检测分子标记物的可能性。方法应用巢式RT-PCR技术检测60例非小细胞肺癌患者外周血中靶基因的表达,并以15例肺良性疾病及20例健康人外周血作为对照。结果CK19mRNA、MUC-1mRNA及LUNXmR-NA表达的阳性率为:51.7%(31/60)、80.0%(48/60)和55.0%(33/60)。对照组中15例肺良性疾病及20例健康人外周血中无LUNXmRNA表达,20例健康人外周血中有1例CK19mRNA表达,而MUC-1mRNA表达的阳性率在肺良性疾病中为60.0%(9/15),在健康人外周血中为65.0%(13/20)。结论CK19mRNA、LUNXmRNA作为检测非小细胞肺癌患者外周血微转移的分子标记物,具有良好的敏感性和特异性,而MUC-1mRNA作为诊断非小细胞肺癌微转移的指标还需进一步探讨。 Objective : Using nested reverse transcriptase polymerase chain reaction (Nested RT-PCR)with primers specific for CK19, MUC- 1 and LUNX to detect circulating tumor cells in peripheral blood of non-small cell lung cancer(NSCLC) patients, and to find cut the relationship between the mRNA expression and micrometastases. Methods The expression of CK19 mRNA, MUC-1 mRNA and LUNX mRNA were analyzed with Nested RT-PCR in 60 NSCLC in peripheral blood, compared with 15 patients with benign lung disease and 20 healthy volunteers. Results The Expressions of CK19 mRNA,MUC-1 mRNA and LUNX mRNA were 51.7% (31/60), 80.0% (48/60) and 55.0% (33/60) in peripheral blood of 60 NSCLC patients. In the control group, there was no expression of LUNX mRNA in 15 patients with benign lung disease and 20 healthy volunteers. One healthy volunteer expression CK19 mRNA. while the expressions of MUC-1 mRNA in 15 patients with benign lung disease and 20 healthy volunteers were 60.0% (9/15) and 65.0% (13/20). Conclusion CK19 mRNA and LUNX mRNA might be specific and sensitive markers to detect circulating tumor cells in peripheral blood of NSCLC patients, while the value of MUC-1 as the marker of micrometastases in peripheral blood is not sure.
