摘要
目的:为了解4种皮肤提取物(HNSE、HBSE和RNSE、RBSE)对EC功能与形态结构的影响。方法:采用人与大鼠肺微血管EC培养模型。动态观察人与大鼠两种烧伤皮肤提取物四种不同浓度作用于相应肺EC后培养液中ET-1、NO及TNF-α体液因子的检测和镜下细胞形态学的变化。结果:(1)用两种烧伤皮肤提取物滴定培养的EC后,其三种因子均异常升高,与相应的对照组及新鲜皮肤提取物组有显著或非常显著的差异(P<0.05或<0.01)。(2)EC形态学观察人或大鼠烧伤皮肤提取物(HBSE、RBSE)均以1000μg及2500μg组其细胞形态学结构出现异常变化明显。结论:人与大鼠烧伤皮肤提取物作用体外EC培养会导致其EC培养液中三种因子的异常变化及结构改变。
Objective:To investigate the effects of normal and burned skin extracts on the function and structure of endothelial cells in vitro.Methods: Four gradient concentrations of extract from burned skin of both human and rat were used in the pulmonary vascular endothelial cell culture, repectively. The supernatants of the cell culture were tested for ET 1, NO and TNF α. The morphological changes of the cultured cells were closely observed as well.Results: (1) A Significant elevation of ET 1,NO and TNF α was observed in cells cultured with burned skin extracts as compared with sham and normal skin extract controls. (2)Significant structural alterations occured in the cells cultured with burned skin extracts in the concentrations of 1 000 μg/L and 2 500 μg/L. Conclusion: The culture of endothelial cells with burned skin extracts of both human and rat may lead to significant morphological changes in cultured cells and their increasing release of certain cytokines and molecules as well.
