摘要
用卵巢癌细胞系A2780在体外分别与化疗药物长春新碱(VCR)、顺铂(DDP)和阿霉素(ADM)作用,获得A2780/VCR、A2780/DDP和A2780/ADM3个耐药细胞亚株,通过体外细胞毒性实验(MTT法),观察A2780与药物作用后的耐受情况及耐药细胞株对10种化疗药物的敏感性。结果:三株细胞对VCR、DDP和ADM均有耐药,对10种作用机制不同的化疗药的耐药指数相差很大。提示:三株卵巢癌细胞均有多药抗药性(MDR),药物引起的耐受可分为两类型:单向性交叉耐药和双向性交叉耐药。
Anticancer drugs VCR,DDP and ADM were introduced respectively into cuetures of ovarian cancer cell line A2780.Three drug resistant subcell lines,A2780/VCR,A2780/DDP and A2780/ADM,were cultured.10 kinds of anticancer drugs,including the above mentioned three drugs,were introduced respectively into these three subcell lines and A2780 cell line,and were examined by MTT method.The results showed all subcell lines not only were resistant to VCR,DDP and ADM respectively,but also resistant to other anticance drugs.The investigation showed that these three ovarian cancer subcell lines all possess multiple drug resistance.The resistance produced by drugs can be classified into two type:single direction cross drug resistance and dual direction cross drug resistance.
