摘要
目的测定都江堰市不同树龄厚朴根皮中所含厚朴酚、和厚朴酚含量,试图为厚朴质量评价、遗传改良及合理采收提供理论依据。方法应用高效液相色谱法测定,色谱柱为Nucleosil C18(5μm)键合相填充柱;流动相为甲醇-水(78∶22);流速为1 ml·min^-1,检测波长为294 nm,柱温为20℃。结果厚朴酚含量在树龄25年时达到最大值为9.2%,和厚朴酚含量在树龄30年左右时达到最大值为7.96%。结论根皮中厚朴酚、和厚朴酚的含量均与植物的生长发育年龄有关。厚朴药材的最佳采收期应在20-30年间。
Objective To determine the contents of magnolol and honokiol from different ages of Magnolia officinalis roots in Dujiangyan by HPLC method and to provide a theoretic basic for quality assessment, genetic improvement and rational picking of magnolia officinalis. Methods Nucleosil C18 (5μm, 250 mm ×4.6 mm) was used. The mobile phase consisted of methanol, water (78: 22). The detection wavelength was 294 nm. The temperature of column was 20℃. Results The content of magnolol was 9.2% when the tree was 25 years old, the content of honokiol was 7.96% when the tree was 30 years old. Conclusion The results show that the contents of magnolol and honokiol relate with the age of the tree. The best time to reap is 20 to 30 ages of the tree.
