摘要
目的:研究叶绿素显示液(CDS)对小鼠直肠淋巴结的染色作用,为其在医学科研和临床中的推广应用提供实验依据和理论参考。方法:24只成年鼠随机分为LD1组和LD2组。LD2组小鼠直肠壁内注入叶绿素显示液(CDS)。两组皆清点直肠各级淋巴结个数,LD1组取正常淋巴结,LD2组取显色淋巴结,石蜡切片,光、电镜观察。结果:于直肠壁内注射CDS,淋巴结染色迅速,不易沉淀,各级哨兵淋巴结(SLN)在30min内被染成翠绿色,与周围组织对比明显;LD2组平均每鼠检出淋巴结6.92个,显著高于LD1组(P<0.01);染色淋巴结24h后仍无褪色,且染色后细胞超微结构无明显改变;术中和术后均未发现不良反应和并发症。结论:①CDS是一种优良的淋巴显色剂,染色迅速,不易沉淀,显色淋巴结与周围组织对比明显,染色持久,且染色后淋巴结细胞超微结构无明显改变,术中和术后无不良反应发生。②LD2组CDS注射后平均每只小鼠观测到淋巴结数目显著高于LD1组,提示CDS在直肠癌根治术中可染色淋巴结,指导淋巴结清扫。
Objective： To study the effect of chlorophyll disclosing solution（CDS） staining lymphonodi rectales of mice, in order to provide reference of experiment and theory in scientific research and clinical study in the future.Methods：Twenty-four mice were divided into LD1 and LD2 group randomly. CDS was injected into rectal wall of the mice in LD2. The number of lymphonodi rectales was counted in two groups. We collected lymph nodes of rectal in group LD1. And then we took out of the stainning lymph nodes in LD2, the hematoxylineosln was performed.Results：By chlorophyll disclosing solution coloring lymph nodes, we found, by injecting the solution into rectal wall of mouse, it colored lymph nodes quickly（in 30 minutes） and was not easy to precipitate. The dyeing lymph nodes showed emerald green, it contrasted sharply with surrounding tissues and no color fading after injected 24 hours. The average number of lymph nodes observed in each mouse was 6.92, which was more than that of LD1 group significantly （P〈0.01）. No adverse reaction or complication were observed in the course of injection and after injection, And the ultrastructure of the lymph nodes colored at different interval was no significant difference to the ones uncolored.Conclusion：The chlorophyll disclosing solution is an ideal dye for staining lymph nodes, which colores them quickly and not easily to precipitate, and the dyeing lymph nodes contrasted sharply with surrounding tissues, and no toxic symptoms or complication is observed after staining, the uhrastructure of the lymph nodes colored at different interval is no significant difference to the ones uncolored. As shows that the chlorophyll disclosing solution can stain lymph system and guide clearance of lymph nodes in radical resection of rectal carcinoma.
