HMME-PDT对兔耳增生性瘢痕块成纤维细胞增殖能力及其超微结构的影响 被引量:7

摘要 目的观察血卟啉单甲醚(hematoporphyrin monomethyl ether,HMME)-光动力学疗法(photodynamie therapy,PDT)对兔耳增生性瘢痕块中成纤维细胞增殖能力及细胞超微结构的影响。方法建立兔耳增生性瘢痕模型后,将24个瘢痕块随机分为HMME-PDT治疗组和对照组(n=12)。造模后28 d取材行银染色,计算瘢痕成纤维细胞核内AgNORs颗粒数,并在透射电镜下观察细胞超微结构改变,分析HMME-PDT对瘢痕的影响。结果HMME-PDT治疗后,银染成纤维细胞核内AgNORs颗粒数显著下降,细胞内粗面内质网萎缩且数量减少;核糖体脱颗粒、解聚;线粒体肿胀,嵴溶解;并出现部分细胞凋亡现象。结论在瘢痕形成早期,HMME-PDT能够抑制兔耳瘢痕成纤维细胞的增殖能力,破坏前胶原蛋白合成的场所,减少胶原蛋白的合成与分泌,并能诱导部分成纤维细胞进入凋亡途径,HMME-PDT有可能成为瘢痕防治的有效方法。 Objective To investigate the effect of hematoporphyrin monomethyl ether-photodynamic therapy( HMME-PDT ) on hypertrophic scar fibroblasts(HSF) in the rabbit ear. Methods The acute model of dermal hypertrophic scar in the rabbit ear was established. Twenty four scar wounds were randomly divided into HMME-PDT treatment group and control group (n = 12). Specimens were harvested from scar wounds on postoperative day 28. The mean AgNORs count of dermal fibroblastic cells in each tissue was determined through standard silver-staining method. Meanwhile, HSF was observed by transmission electron microscopy (TEM).Results In control group, the mean AgNORs count of dermal fibroblasts was 3.01 ± 0. 64, whereas PDT group had mean AgNORs count of 1.87 ± 0. 72, respectively. Mitochondria, rough ER and Golgi of HSF revealed abundant in control group, while decreased and destroyed by PDT. Some fibroblasts underwent marked apoptosis after HMME-PDT. Conclusions HMME-PDT seemed to have good potent effects to inhibit HSF's proliferation,and collagen's synthesis.
