姜黄素对缺血性脑损伤炎症反应的影响 被引量:1

摘要 目的探讨姜黄素(Curumin)对大鼠缺血性脑损伤后白介素1β(IL-1β)和肿瘤坏死因子α(TNFα)表达的影响。方法采用线栓法阻塞大鼠大脑中动脉(MCAO),建立大鼠局灶性脑缺血模型,观察不同浓度姜黄素(100mg/kg、200mg/kg)灌胃给药对缺血性脑损伤后24h大鼠神经学评分、脑梗死体积的改变,并检测血浆IL-1β和TNFα的水平。结果姜黄素两个剂量组与模型组相比,前者行为学评分优于后者(均P<0.05);姜黄素两个剂量组脑梗死体积比模型组明显缩小(均P<0.05);模型组与假手术组相比,血浆IL-1β和TNF-α的水平显著升高,姜黄素两个剂量组与模型组相比,血浆IL-1β和TNFα的水平明显降低(均P<0.05)。结论炎症反应参与脑缺血损伤,姜黄素可以显著降低血浆IL-β和TNFα的水平,对大鼠缺血性脑损伤有明显的保护作用。 Objective To explore the protective effect of curcumin on the cerebral ischemia injury in rats. Methods The focal cerebral ischemia injury models were made by suture - occluded method. Curcumin of 100 mg/kg and 200 mg/kg was given by lavage respectively, and the changes of the neurologic deficiency scores and the infarct volume were observed. The levels of interleukin - 1β( IL - 1β ) and tumor necrosis factor - α(TNFα) in plasma were measured following ischemia 24h. Results Compared with the ischemia group, the two curcumin groups got better score behavior obstacle ( all P 〈 0. 05 ). The infarct size of cerebra was markedly decreased than that of the ischemia group ( all P 〈 0. 05 ). Compared with the vehicle group, the levels of IL - 113 and TNFα in ischemia group increased, administration of curcumin decreased the level of IL - 1β and TNFα in plasma ( all P 〈 0.05 ). Conclusion Inflammatory response plays important role in cerebral ischemic injury in rats. Curcumin can reduce the levels of IL - 1β and TNFα, which can provide protective effect on cerebral ischemic injury in rats.
