Expression and clinical significance of heat-shock protein 90α in gastric cancer tissues

摘要 目的观察热休克蛋白90α(HSP90α)在人胃癌组织和正常胃黏膜组织中的表达,探讨其表达与胃癌的发生、发展及生物学行为的关系。方法收集46例胃癌组织和相应的正常胃黏膜组织标本,分别应用免疫组织化学技术中链霉菌抗生物素蛋白-过氧化物酶连接法(即SP法)和逆转录-聚合酶链反应(RT-PCR)法检测HSP90α的表达情况,分析检测结果。结果HSP90α在胃癌组织中表达的阳性率为82.6%(38/46),明显高于正常胃黏膜组织中的表达(10/46,21.7%);在不同分化类型的胃癌组织中,各组间HSP90α阳性表达率差异无统计学意义(P〉0.05);在淋巴结转移为N1期以后(不包括N1在内)的病例中HSP90α表达阳性率为96.2%(25/26),高于淋巴结转移为N0和N1的病例;在TNM分期Ⅲ、Ⅳ期阳性表达率为92.9%(26/28),也要高于Ⅰ、Ⅱ期。结论HSP90α在胃癌组织中呈现高表达,并且在淋巴结转移多和越晚期的胃癌组织中表达越高,提示HSP90α可作为胃癌预后判定监测指标。 Objective To investigate the expression of heat-shock protein 90α (HSP90α) in gastric cancer tissues and normal gastric mucosa, and to find out its relationships to the carcinogenesis, developing and biological behavior of gastric carcinoma. Methods The specimens of 46 patients with gastric cancer were collected, including 46 cases of gastric carcinoma and normal mucosa. Immunohistochemical staining and RT-PCR were performed in these tissues for the expression of HSP90α, and the results were analyzed. Results The positive expression rate of HSP90α in gastric cancer tissues was 82.6% ( 38/ 46 ) , which was higher than in normal mucosa (21.7% , 10/46 ). There was no significant difference in the expression of HSP90α among different differentiation types of gastric cancer. The positive expression rate of HSP90α in gastric cancer in N2 ,N3 stages was 96.2% (25/26) ,higher than that in gastric cancer in NO and N1 stages. The positive expression rate of HSP90α in TNM Ⅲ, stages was 92.9% (26/28) , higher than that in TNM I and II stages. Conclusion The level of HSP90α expression in gastric carcinoma is higher than in normal gastric mucosa. The more the invasive lymph nodes and the more advanced the gastric carcinoma,the more the HSP90α expression. It is suggested that high expression of HSP90α in gastric carcinoma indicates a poor prognosis.
