蛋白酶类测定底物的替代研究

摘要 用0.06M盐酸溶解抗凝猪全血制成1∶8、1∶10血红蛋白液,替代氯化血红蛋白,用市售部标级多酶片为参照物,以国际上公认的胃蛋白酶测定方法之一DAB9法为依托进行底物替代研究试验。结果显示替代后测定的重现性好,相对偏差:多酶片为3.46%,仔猪混合胃液为:A样1.11%,B样0.53%,C样1.90%所测的多酶片含量为48.68单位/片,与经某权威检测部门测定的46.5单位/片相近。相对误差为4.69%。 By using 0.06M hydrochloric acid to dissolve the antifreezing pig blood, two kinds of hemoglobin liquid of 1∶8 and 1∶10 are formed, and serve as the substituent of the hemin. Using the multienzyme complex of ministenal standard as a reference and depending on the international established determining method of the pepase-DAB 9, a test of substrate substitution is done. The results show that the repealability of determining substrates is fine and the relative deviation of the multienzyme complex is 3.46% and the relative deviation of the gastric juics of piglets which include sample A, sample B and sample C is respectively 1.11%, 0.53% and 1.90% and the determining content of the multienzyme complex approaches the content determined by an authentic department. The relative error of the test is 4.69%.
