摘要
目的研究吗啡对体外培养的人正常子宫平滑肌细胞雌激素受体信使核糖核酸(estrogen receptor mRNA,ERmRNA)表达的影响。方法体外培养人正常子宫平滑肌细胞,免疫细胞化学的方法进行鉴定,然后将培养细胞随机分为5组:空白对照组,1×10-7mol/L吗啡组,1×10-6mol/L吗啡组,1×10-5mol/L吗啡组,1×10-4mol/L吗啡组。通过逆转录聚合酶链式反应(RT-PCR)法测定各实验组子宫平滑肌细胞ERmRNA的表达水平。结果不同浓度的吗啡组与对照组平滑肌细胞ERmRNA表达相比差异有统计学意义(P<0.05);在吗啡组平滑肌细胞ERmRNA的表达呈剂量依赖性降低,细胞mRNA表达量与吗啡浓度之间存在负相关。结论外源性阿片类药物吗啡可抑制平滑肌细胞雌激素受体ERmRNA的表达。
Objective To explore the effects of morphine on ER(estrogen receptor)mRNA expression in human myometrial .smooth muscle cells. Methods Human myometrial smooth muscle cells were isolated and cultured in vitro, and identified by immunocytochemical staining. All cells were randomly divided into 5 groups:blank control group, 1 × 10^-7 mol/L morphine group, 1× 10^-6 mol/ L morphine group, 1 × 10^-5 mol/L morphine group, 1 × 10^-4 mol/L morphine group. The expression of ER mRNA was detected by RT-PCR method. Results ER mRNA expression was significantly lower in different morphine groups than in blank control group( P 〈 0.05). ER mRNA expression decreased in a dose dependent manner in morphine groups ( r = - 0. 979, P 〈 0.05 ). Conclusion Exogenous opiate like substance morphine could inhibit the expression of ER mRNA in human myometfial smooth muscle cells.
