摘要
目的探讨血管内皮生长因子(VEGF)、P物质(SP)在大鼠门静脉高压性胃病(PHG)发病机制中的作用。方法雄性SD大鼠随机分为门静脉高压胃病(portal hypertensive gastropathy,PHG)组(n=15)和对照组(n=10)。PHG组大鼠采用门静脉两步法结扎加左肾上腺静脉结扎手术,于门静脉完全结扎后第14天,用计算机辅助多道生理记录仪测量门静脉压力。采用免疫组织化学SP法检测胃黏膜组织VEGF、SP表达情况,对照组测量方法同上。结果实验组大鼠门静脉压力[(13.3±2.29)mmHg]显著高于对照组[(4.4±0.84)mmHg,P<0.05]。免疫组化发现VEGF,SP在实验组大鼠胃黏膜组织中呈强阳性表达,在对照组则呈弱阳性表达。结论VEGF、SP可能参与门静脉高压大鼠胃黏膜局部病变。
Objective To explore the expression of vascular endothelial growth factor(VEGF) ,substance P(SP)in gastric mucosa of portal hypertensive rats. Methods Male SD rats were randomly divided into two groups: portal hypertensive gastropathy(PHG) group( n = 15) and control group( n = 10). A two-stage ligation of portal vein in the rats in PHG group was performed. After two weeks,portal venous pressure was measured with the computer auxiliary multi-channel physiological recorder. The expression of VEGF and SP in gastric mucosa was detected by immunohistochemical Strephavidin-Peroxidase technique. Results The portal venous pressure in PHG group was significantly higher than in control group(P〈0.05). The expression of VEGF and SP in gastric mucosa was stronger in PHG group than in control group. Conclusion VEGF and SP may play a role in the gastric mucosal lesion of portal hypertensive rats.
