Expressive activities and their implications of the key target genes in p38MAPK signaling pathway and the factor of TNF-α in the tissues of nasal polyps

摘要 目的探讨p38MAPK信号传导通路关键靶点基因及TNF-α在鼻息肉发病机制中的介导与调控作用。方法35例鼻息肉标本及15例下鼻甲组织标本,常规HE染色鉴别嗜酸性粒细胞浸润情况,免疫组织化学S-P法检测p38MAPK、P-p38MAPK、TNF-α的活性表达程度,并进行图像分析。结果鼻息肉组中p38MAPK、P-p38MAPK及TNF-α的表达活性均明显高于下鼻甲组(P<0.05),而且p38MAPK及TNF-α表达于胞浆中,P-p38MAPK主要表达于胞核;p38MAPK、P-p38MAPK及p38MAPK和TNF-α蛋白的表达具有相关性。在对照标本,没有或仅少量表达p38MAPK、P-p38MAPK及TNF-α。结论在鼻息肉的发病机制中,p38MAPK信号传导通路处于激活状态,并且可能通过TNF-α蛋白表达活性的增强而加剧黏膜炎症反应,促进鼻息肉的发生与发展。 Objective To investigate the mediating and regulating effects of the key targeting genes in p38MAPK signaling pathway and the factor of TNF-α on the pathogenesis of nasal polyps in the developing process of this lesion. Methods Included in this study were 35 samples of nasal polyps and 15 samples of inferior turbinate, with routine HE staining procedures to identify the infiltrating status of eosinophils (EOS) and S-P immunohistochemical procedures to determine the expressive activities of the key targeting genes, like p38MAPK, P-p38MAPK and TNF-α, in these tissue samples. Following these procedures, an analysis was done on the difference and their implications in the pathogenesis of nasal polyps, with image analyzing techniques to supplement such an analytic program. Results As determined in this trial, the expressive activities of p38MAPK, P-p38MAPK and TNF-α were significantly higher in the tissue samples of nasal polyps than that in inferior turbinate (P〈0.05), with the significant expressive activities of p38MAPK and TNF-α displayed in the cytoplasm and that of P-p38MAPK in nuclei of these samples. Furthermore, there was market correlating tendency present as shown in the expressive activities among p38MAPK with P-p38MAPK and p38MAPK with TNF-α in these studied samples. In contrast, the expressive activities of p38MAPK, P-p38MAPK and TNF-α were very lower, even no significant expressive activities could be determined in the controlling samples of inferior turbinate. Conclusions It could be concluded from the data as shown in this study that, in the pathogenesis of nasal polyps, the signaling pathway of p38MAPK is in an activated way so as to promote the inflammatory responsive process in diseased nasal mucosa via the route of elevated TNF-α expressive activity. As a result of such a pathological event, aggravated should be the initiating and developing process of this condition.
