摘要
本课题主要研究T细胞受体(TCR)识别抗原后Vβ基因发生重排,mRNA表达水平改变。用紫外线处理的单纯疱疹病毒-2型(HSV-2)、HSV-2及PHA分别感染或刺激正常人的PBLs,培养4~6天后,提取mRNA,并采用RT-PCR、Southern杂交发现识别HSV-2的TCRVβ2、6、7、8基因在体外选择性扩增。在采用HSV-2攻击皮肤病患者发作期、缓解期以及再发作期的PBLs时,发现TCRVβ基因表达随着病程变化而改变,尤其Vβ7亚家族表达水平显著高于其它亚家族基因,这显示了TCRVβ基因的变化是恒定特异性的。
Human peripheral blood lymphocytes(PBLs) of healthy people and patients were infected or stimulated with herpes simplex virus(HSV 2),ultraviolet radiated HSV 2(HSV 2 UV) and PHA,respectively and cultured separately for 4 to 6 days.The mRNAs from the PBLs were extracted separately and the expression level of T cell receptor (TCR) Vβ gene were determined.The results showed that TCR Vβ6,7 and 8 genes were selectively amplified.The expression levels of different genes of TCR Vβ varied at different clinic stages when stimulated with HSV.It suggests that the expression of TCR Vβ genes is of absolute specificity.
