摘要
目的 探讨高效液相色谱法(HPLC)测定酮洛芬粉针剂中酮洛芬的含量。方法 采用美国Agilent C18色谱柱(200mm×4.6mm,5μm),以0.01mol.L^-1磷酸二氢钾溶液(用磷酸调pH至3.5)-甲醇(55∶45)为流动相,流速为1.0mL.min^-1,检测波长为256nm。结果 酮洛芬在0.005125-0.05125mg.mL^-1范围内呈良好的线性关系,r=0.9996(n=6);平均回收率为99.93%,RSD为0.17%(n=5)。结论该方法结果准确,精密度和重现性好,可用于酮洛芬粉针剂的含量测定。
OBJECTIVE To establish an HPLC method for content determination of ketoprofen powder for injection. METHODS The HPLC analysis was carried out using a column of Agilent Cls(200mm × 4.6mm, 5um), a mobile phase consisted of 0.01 mol·L^-1 KDP solution (pH3.5)-methanol (55:45), at a flow rate of 1.0 mL· min^-1 and detection at 256nm. RESULTS The calibration curve was linear in the range of 0. 005125-0. 05125 mg·mL^- 1(r=0. 9996, n= 6). The mean recovery rate was 99.93 %, with a RSD of 0.17% (n = 5). CONCLUSION The method is simple, accurate and reliable.
