应用6-DMAP人工诱导近江牡蛎三倍体的初步研究 被引量:4

摘要 采用6-二甲基氨基嘌呤(6-DMAP)抑制受精卵第二极体排放法诱导近江牡蛎产生三倍体,通过6-DMAP浓度、诱导时机、诱导持续时间3因素3水平正交试验设计,得出近江牡蛎三倍体诱导3因素最优水平组合,即当30%受精卵出现第一极体时,将受精卵浸泡在含6-DMAP 450μmol/L的海水中20 min;3因素影响诱导率的主次顺序为:6-DMAP浓度→诱导时机→诱导持续时间。 It was undertaken to induce triploid of Crassostrea rivularis by blocking the second polar body of the zygotes with 6-Dimethylamin- opurine (6-DMAP). L9 (3^4) design was selected to engage in orthogonal experiment of three factors and three levels:three concentrations of 6-DMAP were 300,450 and 600 μmol/L;three levels of induced occasion (the percentage of zygotes with polar body I when observing the zygotes in still) were 10 % ,30 % and 50 % ;and three levels of induced duration were 10,15,20 min, respectively. Sampling was repeated twice. Sperms and ovums were obtained with anatomy method, ripen in vitro. Ovums were washed and inseminated artificially. Ploidy was detected by flow cytometry Partec I made in Germany. According to results of orthogonal test and intuitive analysis of triploidy, the optimal conditions of treatment is the 6-DMAP concentration of 450 μmol/L, the induced occasion when 30 % of zygotes presenting polar body I , and the duration of 20 min. The sequence of three decisive factors is as follows:the concentration of 6-DMAP,induced occasion, and induced duration.
