摘要
选用草原红牛30头、蒙古牛10头、夏洛来牛10头、利木赞牛6头和西门塔尔牛10头组成的试验牛群体共计66头,经过牛血液或肝脏基因组DNA的提取、8对微卫星引物的PCR扩增、扩增产物的聚丙烯酰胺电泳分型、分析各位点等位基因及全部个体的标记基因型、PPAP3.0和SPSS10.0软件计算基因频率、多态信息含量(PIC)和杂合度、遗传距离等步骤从分子水平上分析了在8个位点的遗传多态性。结果表明,微卫星位点IDVGA2、IDVGA46、TGLA44、BM1824、ETH225、BM2113、IDVGA44、IDVGA55扩增出的条带数分别为6、6、6、5、4、2、6、4条,平均条带数分别为4.4、4、4.2、3.6、3.4、2、4.8、3.2条,平均PIC/H分别为0.683 1/0.733 7、0.596 3/0.660 2、0.646 2/0.700 1、0.558 1/0.630 2、0.552 9/0.615 8、0.371 1/0.417 4、0.683 1/0.728 4、0.5431/0.615 3,均属于高度多态性位点。用于群体遗传标记的研究是较理想的。
In this study, a group of Charolais, Limousin, Simmental, Steppe Red Cattle, Mongolian Cattle was chosen and its population genetic polymorphism was analyzed in the level of molecule by the process of genome DNA extraction from blood, PCR amplification of 8 primers, fragment typing with polyacrylamide gel electrophoresis (PAGE), allele and genotype analysis of every locus by UVIBAND system, calculation of allele frequencies, Polymorphism information content (PIC) and Heterozygosity by PPAP3.0 and SPSS10.0. The results showed that the number of alleles in IDVGA2, IDVGA46, TGLA44, BM1824, ETH225, BM2113, IDVGA44, IDVGA55 was 6, 6, 6, 5, 4, 2, 6, 4, and the polymorphism information content (PIC) and H was 0.6831/0.7337, 0.5963/0.6602, 0.6462/0.7001, 0.5581/0.6302, 0.5529/0.6158, 0.3711/0.4174, 0.6831/0.7284, 0.5431/0.6153. All of them were high polymorphic loci. So they were ideal loci in the study of looking for genetic markers related to the production performance.
