摘要
宫颈癌细胞(HELA)、人胚肾细胞(HEK293)和中华仓鼠卵巢细胞(CHO)3种细胞系在用于哺乳动物细胞悬浮培养的锥底生物反应器(FCB)、平底生物反应器(FB)和常规机械搅拌式生物反应器(STR)中进行培养和冷模实验。结果表明:FCB中HELA细胞的最高密度比FB高27%,HEK293和CHO细胞的最高密度分别比STR高38%和43%。FCB的混合时间平均比FB短27%,比STR长35%;kLa比FB和STR分别高83%和97%。
Three cell lines of HELA, HEK293 and CHO were cultured in a novel inverted frusto-conical bottom bioreactor (FCB), flat bottom bioreactor (FB) and conventional mechanism stirred bioreactor (STR), respectively. Highest cell densities in FCB for HELA,HEK293 and CHO cells were 27% ,38% and 43% higher than that in FB,STR and STR, respectively. Additionally, mixing time and ki.a of three types of bioreactors were determined. Average mixing time of FCB is 27% shorter than that of flat bottom one, while 35% longer than that of STR. Average kLa of FCB is 83% and 97% higher than that of FB and STR, respectively.
