摘要
为了探明家蚕对浓核病毒(镇江株)抵抗性和感受性品种间在分子机制上的差异,通过蛋白质双向电泳(2-DE)和基质辅助质量飞行时间质谱技术(MALDI-TOF MS)对两个不同抗性家蚕品种的中肠组织蛋白进行了比较分析。2-DE电泳结果表明,两个品种间的中肠组织蛋白斑点数及位置、形态等差异很小,进一步比较获得了9个差异蛋白斑点,其中感受性蚕品种JS有6个,抵抗性蚕品种NIL有3个。经MALDI-TOF MS鉴定结果显示,感受性品种JS有4个差异蛋白斑点可能分别为氢离子转运ATP合酶β亚基1、氢离子转运ATP合酶β亚基2、组织蛋白酶D或3-羟酰辅酶A脱氢酶;抵抗性品种NIL中有2个差异蛋白点可能是组织蛋白酶。
To investigate the difference of proteins in the midgut of silkworm larva of strains susceptible (JS) and non-susceptible (NIL) to Bombyx mori densovirus (Zhenjiang Strain, BmDNV-ZJ), the proteins extracted from the midgut were analyzed comparatively by two-dimensional electrophoresis (2-DE) and matrix-assisted laser ionization time-of-flight mass spectrometry (MALDI-TOF MS). The results showed that there was less difference in the number, location and shape of the protein spots on the 2-DE image between two strains, and 9 specific spots, 6 in JS strain and 3 in NIL strain, were identified. MALDI-TOF MS analysis of the specific spots indicated that 4 specific spots of JS strain belonged to H ~ transporting ATP synthase beta subunit isoform1 and 2, cathepsin D, and 3-hydroxyacyI-CoA dehydrogenase, respectively. And 2 specific spots of NIL strain might be cathepsin.
