摘要
目的研究比较大鼠脑梗死(MCAO)、出血性脑梗死(HI)模型脑组织含水量及Na+-K+-ATP酶活性的变化。方法54只SD大鼠随机分为3组:假手术组、MCAO组和HI组。采用干-湿重法测定脑组织含水量,无机磷法测定Na+-K+-ATP酶活性。结果HI组脑组织含水量在缺血6h、MCAO组缺血12h后较假手术组间差异均有统计学意义(P<0.01),缺血6h和12h时MCAO组较HI组差异有统计学意义(P<0.05)。MCAO组和HI组Na+-K+-ATP酶活性较假手术组间差异有统计学意义(P<0.01),缺血3~24h时MCAO组较HI组间差异有统计学意义(P<0.05)。结论Na+,K+-ATP酶活性降低及其引发的脑水肿可加重原有的脑梗死。
Objective To study the changes of brain water content and activity of Na^+ - K^+ - ATPase in rats with middle cerebral artery occlusion (MCAO) and hemorrhagic cerebral infarction (HI). Methods Totally 54 Sprague - Dawley rats were randomly divided into sham - operated, MCAO and HI groups. Wet and dry weight methods were used to measure the changes of brain water content. The method of spectrophotometry was applied to measure the activities of Na ^+ - K ^+ - ATPase. Results The brain water contents of rats in the HI group 6 h after ischemia and in the MCAO group 12 h after ischemia were significantly higher than that in the sham group ( P 〈 0. 01 ). The brain water content of rats in the HI group was higher than that in the MCAO group ( P 〈 0. 05). The brain water content of rats in the MCAO group after 6 h and 12 h ischemia were higher than that in the HI group ( P 〈0. 05). The Na^+ - K^+ - ATPase activities of rats in the operated groups ( MCAO and HI) decreased significantly as compared with that in the sham group ( P 〈 0. 01). The Na ^+ - K ^+ - ATPase activity of rats in the HI group was lower in than that in the MCAO group from 3 to 24 h ( P 〈 0. 05). Conclusion The decrease of the Na ^+ - K ^+ - ATPase activity and its inducing edema may make the cerebral infarction deteriorated.
