摘要
目的验证发酵莲子乳(Lotus-milk Fermented Production,LMFP)保护小鼠肠粘膜屏障功效,并探讨可能的作用机制。方法将60只清洁级ICR小鼠随机分为5组:阴性对照组和模型组给予蒸馏水,发酵莲子乳组、莲子匀浆组、发酵乳组,分别给予相应干预物。未次灌胃后腹腔注射细菌脂多糖(LPS)6 mg/(kg.bw),阴性对照组给予等体积/体重蒸馏水,18 h后处死小鼠,采集样本检测指标。结果与阴性对照组比较,各组小鼠均出现不同程度的萎靡症状,甚至死亡,其中模型组最严重,发酵莲子乳组症状最轻;与模型组比较,发酵莲子乳可明显升高小鼠外周血白细胞总数和肠道分泌型免疫球蛋白A(SIgA)含量(P<0.001,P=0.001),有效维持肠道内乳酸杆菌菌量(P<0.001),降低LPS所升高的血浆肿瘤坏死因子-α(NF-α)水平(P=0.01),抑制大肠埃希菌过度繁殖(P=0.001),且发酵莲子乳功效优于发酵乳,而莲子匀浆无保护作用。结论利用LPS腹腔注射小鼠成功建立肠粘膜损伤屏障模型,发酵莲子乳可通过维持肠道菌群稳定,增强肠道免疫机能等方式修复肠粘膜屏障损伤,减轻全身炎症反应程度。
Objective To explore the effect and mechanism of lotus- seed milk fermented production(LMFP) on the intestinal mucosaI barrier of mice. Methods 60 male ICR mice were randomly assigned into five groups. The mice were pretreated intragastrically with distilled water, lotus - seed milk fermented production, lotus - seed milk and fermented milk once daily for two weeks. Twenty - four hours after the last dosing, animals were administered with LPS [ 6 mg/( kg·bw)] intraperitoneally, control animals were given distilled water only. Eighteen hours after the intoxication, the indices were measured. Results The mice injected with LPS showed the symptoms of sluggish in varying extent, especially in model group. The LMFP group was the most slight compared with other groups. The lotus - seed milk fermented production can significantly improve the concentration of intestinal SIgA and the quantity of white blood cell( P 〈 0. 001, P = 0. 001 ), retain the amount of lactobacillus acidophilus( P 〈 0. 001 ), decrease the concentration of TNF - α in serum induced by LPS and the quantity of Escherichia coli in the mouse's caecum. The protective effect of lotus- seed milk fermented production on the intestinal mucosal barrier harmed by LPS was significantly better than fermented milk, and there was no protection in lotus - seed milk group, Conclusion The model of intestinal mucosal barrier harmed by LPS was established successfully, The lotus - seed milk fermented production can protect the intestinal mucosal barrier against harm caused by LPS, and relieve the response of systemic inflammatory.
