摘要
目的建立健康家兔服用大川芎丸后血清中阿魏酸(FA)的高效液相色谱测定法(HPLC),并研究阿魏酸在大鼠体内的药代动力学。方法用HPLC法,色谱柱为Inertsil ODS-3(5μm,4.6×250mm),流动相为:甲醇∶水∶冰乙酸(49∶50.745∶0.255,V/V),流速1.0mL·min^-1,检测波长322nm。FA用内标(香豆精)法定量。结果FA的最低检测限为0.25ng,最低血清检测浓度为5ng·mL^-1(S/N〉3),线性范围是26.4-4224ng·mL^-1,r=0.9999.平均加样回收率为98.17%±1.26%.结论该方法灵敏、快速、简便、专一性和重现性好,可用于阿魏酸血药浓度分析及药代动力学研究。
objective To establish the method for determining ferulic acid in serum by HPLC and study the pharmacokinetics in healthy rabbit after taken DCXW.Methods Inertsil ODS-3(5 μm,4.6×250 mm)was used as the stationary phase,methanol-Water-acetic acid(49∶50.745∶0.255,V/V)was used as mobile phase,flow rate of mobile phase was 1.0 mL·min^-1,wavelength of detection was 322 nm.The Coumarin was used as inner standard.Results The detection limit of FA was 0.25 ng,and the lowest detectable concentration of FA was 5 ng·mL^-1,with a linearity of 26.4-4224 ng·mL^-1,r=0.9999.The mean recovery rate of FA was 98.17%±1.26%.Conclusion This method is sensitive,fast,simple,specific and reproducible.It can be used to the analysis of FA in serum and the study of pharmacokinetics.
