摘要
目的探讨补肾富钙中药对大鼠卵巢辐射损伤所致骨质疏松形态计量学的影响。方法40只SD大鼠随机分为四组(n=10),分别为假手术组(灌服1%纤维素90 d)、模型组(以50 Gyγ射线局部照射大鼠卵巢建立卵巢辐射损伤模型,1%纤维素灌服90 d)、尼尔雌醇组(造模后给予尼尔雌醇灌胃90 d)和补肾富钙中药组(造模后给予补肾富钙中药灌胃90 d)。各组大鼠于处死前第14天和第4天分别腹腔注射盐酸四环素(30 mg/kg)进行标记,处死后取右侧股骨组织和L4腰椎组织分别进行骨组织形态计量和扫描电镜观察。结果与模型组比较,补肾富钙中药组和尼尔雌醇组的骨小梁体积显著增加,骨吸收表面减少;尼尔雌醇组的骨形成表面和矿化沉积率下降,但在补肾富钙中药组则无显著变化。扫描电镜观察显示,模型组骨小梁变细、中断、排列稀疏,而补肾富钙中药组和尼尔雌醇组上述变化较轻微。结论补肾富钙中药可有效预防卵巢辐射损伤诱发的骨质疏松。
Objective To explore the effects of Bushenfugai Traditional Chinese Medicine on the morphometry of osteoporosis induced by ovary irradiation in rats. Methods Forty SD rats were randomly divided into four groups with ten in each group. The sham operation group were administered with 1% carboxymethyl cellulose for 90 days. For the model group, the ovaries were irradiated by 50 Gy γ-ray to establish the animal model, and 1% carboxymethyl cellulose was administered for 90 days. For the nilestriol group and Bushenfugai group, intragastric administration of nilestriol and Bushenfugai Traditional Chinese Medicine were performed respectively for 90 days after the establishment of animal model. The labeling was done by intraperitoneal injection of Tetracycline Hydrochloride (30 mg/kg)in each group 14 days and 4 days before sacrifice. After sacrifice, the tissues harvested from right lateral thigh and L4 were subjected to bone histomorphometry and scanning electron microscopy. Results Compared with model group, the volumes of bone trabecula were increased in both Bushenfugai group and nilestriol group with decreased bone absorbing surface. The bone forming surface and rate of mineralization deposition of nilestriol group were decreased, while there were no significant changes in Bushenfugai group. It was revealed by scanning electron microscopy that the bone trabecula of model group was thin, interrupted and sparsate, while those were improved in both Bushenfugai group and nilestriol group. Conclusion Bushenfugai Traditional Chinese Medicine may effectively prevent osteoporosis induced by ovary irradiation.
