木豆种子超干保存最适含水量的选择及其机制分析 被引量:5

Selection of Optimum Moisture Content of Cajanus cajan Seeds for Ultra-dry Storage and Its Mechanism Analysis

摘要 采用硅胶与烘干干燥相结合的方法,将木豆种子脱水至1.17%、2.23%、3.34%、4.03%、4.94%、6.08%和7.12%7个含水量梯度,在常温下保存1a,再结合室内回湿、PEG回湿等处理后,测定其发芽率、脯氨酸(Pro)的质量分数、丙二醛(MDA)质量摩尔浓度、总糖的质量分数和脂肪酸组分。结果表明,木豆种子较为适宜的脱水范围为4.03%-7.12%,在此范围内经过回湿处理,其发芽率都在60%以上,保存效果优于常温保存下的未经脱水的种子(CK1),其中以含水量为4.94%的种子的保存效果最佳,其发芽率高达75%。对Pro的质量分数、MDA的质量分数、总糖的质量分数、脂肪酸组分的测定显示:适宜的脱水和回湿处理使种子的脯氨酸的质量分数、脂肪酸不饱和指数增加,总糖的质量分数降低,种子膜质过氧化减轻,从而使种子保持较高的生活力。 Cajanus cajan seeds were dried to different moisture contents ranging from 1.17% to 7.12% by means of silica gel and firing, and then stored at room temperature for 12 months. The germinability, proline content, malondialdehyde (MDA) content, total sugar content and fatty acid composition of the seeds were studied combined with different treatments of humidification. Results show that the optimum moisture content of C. cajan seeds is ranging from 4.03% to 7.12%. Within this range, the germinability of the seeds is higher than that of the seeds stored at room temperature with normal moisture content, exhibiting a germination rate above 60%. The optimum moisture content of the seeds is 4.94%, with a germination rate over 75%. For the seeds with appropriate moisture content after treatment of humidification, the proline content and the index of unsaturated fatty acids increase, the total sugar content decreases, and the membrane peroxidation reduces, which make the seeds keep higher vigor.
