木通中共有成分木通酸和木通皂苷Stc的检测 被引量:3

Determination of Components Quinatic Acid and Akeboside St_c Commonly Contained in Mutong of Akebia

摘要 应用薄层色谱和高效液相色谱技术,对木通属3种基源木通(五叶木通、三叶木通和白木通)的不同产地、不同生长期共20个样品进行检测分析,从中发现了共有成分木通酸和木通皂苷Stc。建立一种测定共有成分的反相高效液相色谱方法。采用Agilent SB-C18(4.6 mm×250 mm,5μm)色谱柱,以V(乙腈)∶V(水)=65∶35为流动相,进样量20μL,流速1 mL/min,柱温25℃,检测波长203 nm,共有成分在50 min内得到较好分离,回收率符合含量测定要求。运用该方法对木通药材进行共有成分含量测定,结果显示,共有成分在20个样品中含量差别较大。经查阅文献表明,木通酸仅在木通科木通中存在,为木通的专属性成分。此研究结果可为中国药典木通质量标准修订提供参考。 With thin layer chromatography(TLC) and high performance liquid chromatography(HPLC) technique, quinatic acid and akeboside Stc were found for the first time as commonly contained components in the 20 medicine materials of three Mutong species of Akebia (A. quinata, A. trifoliata and A. trifoliata var. austfalls) with different producing area and different growing period. A HPLC method was achieved for determinating the two commonly contained components. The samples were separated well at 25 ℃ in 50 minutes using Agilent SB-Cls (4.6 mm x 250 mm, 5 μm) column with acetonitrile-water( volume ratio is 65:35 ) as mobile phase. The flow speed, detection wavelength and injection volume was set as 1 mL/min, 203 nm and 20 μL, respectively. The recoveries accorded with the stipulation of content determination. The assayings of the two commonly contained components with the method showed that all of the 20 samples contained the two components, but very differently in content. The existence of quinatic acid in other plants has not been reported yet, this is useful for revising the quality standard in Chinese Pharmacopeia for Mutong from Akebia.
