摘要
目的通过对新的DYS651基因座在山西汉族群体的研究,获得该基因座的基因频率分布情况,为群体遗传学、法医学个体识别及亲子鉴定提供计算数据,对其在法医学上的应用进行可行性探讨。方法在Y染色体基因组DNA中查找候选的基因座,用GenBank中的引物序列进行PCR扩增,再用银染方法显带,并对其筛选的等位基因测序,构建等位基因分型标准物;按照ISFG原则命名各等位基因。结果本研究选择了Y染色体基因座DYS651。在120例山西汉族无关男性个体血样中共检出了5个等位基因。其基因多样性为0.7768,在法医学应用方面,其个人识别能力(DP)和非父排出率(PE)均为0.7768。结论新Y染色体基因座DYS651具有较高的遗传多态性,在法医学及人类遗传学方面具有应用价值。
Objective To obtain the allele frequencies of the Y - STR loci DYS651 and discuss the possibility of applying it to forensic science by investigating this Y - STR loci in shanxi Han eth- nic group. Methods The candidate of Y - STR loci was selected from human Y chromosome ge- nome DNA. Tl-,e products of PCR were analyzed with polyacrylamide gel electrophoresis. Each allele of these Y- STR loci was sequenced and the allele ladders were constructed. Alleles of these Y- STR loci were nominated according to recommendations of the International Society of Forensic Genetics. Results A new Y - STR IocusDYS651, had been chosen. Five alleles had been found in 120 unrelated male venous blood in Chinese Han population of Shanxi. Genetic diversity (GD) was 0.7768, and if used in forensic science, both of the power of discrimination (PD) and the power of exclusion (PE) were 0.7768. Conclusion DYS651 has a high level of genetic polymorphism and can be used in forensic science and human genetics research.
