摘要
目的探讨大鼠肝脏缺血再灌注时肝细胞凋亡与肿瘤坏死因子-α(TNF-α)的关系。方法建立大鼠局部肝脏缺血再灌注模型,将72只健康雄性Wistar大鼠随机分为正常组、假手术组和缺血再灌注组,采用放射免疫法测定再灌注后不同时相中血清TNF-α含量,末端脱氧核苷酸转移酶介导的dUTP缺口末端标记法(TUNEL)和透射电镜观察肝细胞凋亡状态。结果再灌注后1、3、6h和24h血清TNF-α含量及肝细胞凋亡指数(HAI)明显高于正常组和假手术组,且均于6h达到高峰,再灌注后各时相TNF-α水平与HAI呈显著正相关。结论肝脏缺血再灌注损伤过程中,TNF-α表达水平异常上调并可能对肝实质细胞凋亡起介导作用。
Objective To investigate the relationship between hepatocellular apoptosis and tumor necrosis factor-alpha (TNF-α) in rats during hepatic ischemia-reperfusion. Methods With a rat model of acute hepatic ischemia-reperfusion, 72 healthy male Wistar rats were randomly divided into 3 groups: normal group, sham-operated group and ischemia-reperfusion group. The concentrations of serum TNF-α was measured by radioimmunity technique and the hepatocellular apoptosis was detected by TdT-mediated dUTP-biotin nick end labeling (TUNEL) and electron microscope. Results The concentrations of serum TNF-α and hepatocellular apoptotic index (HAI) in ischemia-reperfusion group were higher than that in normal and sham-operated groups. Both TNF-α and HAI reached peak at 6 hour after reperfusion. There was a positive correlation between TNF-α and HAI. Conclusion There is an extraordinary up-regulation of the expression of TNF-α after hepatic ischemia - reperfusion and perhaps TNF-α is an inducible factor of hepatocellular apoptosis.
