摘要
目的研究银杏叶提取物(extract of ginkgo biloba,EGB)抵抗肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)诱导的人肾小管上皮细胞凋亡及其作用机制。方法将人肾小管上皮细胞分成4组:对照组,TNF-α组,EGB+TNF-α组,EGB组,各组细胞培养至80%~90%汇合时加入相应药物干预。采用MTT法检测TNF-α对细胞的毒性作用,通过细胞形态学检测(AO/EB染色)和流式细胞仪计数测定细胞凋亡发生情况,Western blot法测定细胞凋亡相关蛋白Bcl-2、Bax的表达。结果TNF-α对人肾小管上皮细胞的毒性作用呈剂量和时间依赖性;TNF-α(10μg/ml)可显著诱导人肾小管上皮细胞凋亡,而EGB(50mg/ml)可明显抑制它的作用,同时Western blot测定显示EGB可显著上调Bcl-2的表达而下调Bax的表达。结论EGB可以显著抑制TNF-α诱导的人肾小管上皮细胞凋亡,此作用可能与其上调抗凋亡蛋白Bcl-2的表达和下调促凋亡蛋白Bax的表达有关。
Objective To investigate the effects of Ginkgo biloba (EGB) extract against renal tubular epithelial cell apoptosis induced by tumor necrosis factor alpha (TNF-α). Methods The human renal tubular epithelial cells were divided into 4 groups: control group, TNF-α group, EGB+TNF-α group and EGB group. When the cells were grown to 80%-90% confluence, the cells were treated with the corresponding protocols. The cytotoxicity of TNF-α was evaluated by MTT assay. Apoptosis was assessed by AO/EB staining and flow cytometry. The expression of Bcl-2 and Bax was detected by Western blot. Results The cytotoxicity of TNF-α to human renal epithelial cells was dose- and time-dependent. The apoptosis of human renal epithelial cells was significantly induced by TNF-α (10 μg/ml), which could be significantly attenuated by EGB (50 mg/ml). Western blot revealed EGB could significantly up-regulate the expression of Bcl-2 and down-regulate the expression of Bax. Conclusion EGB significantly attenuated the apoptosis of human renal epithelial cells induced by TNF-α, which may be associated with the up-regulated expression of Bcl-2 and down-regulated expression of Bax.
