摘要
目的:寻找胃癌SGC-7901细胞株中SP(side population)亚群细胞的特异性标志物。方法:用Hoe-chest33342染色检测SGC-7901细胞株中SP细胞的比例;通过荧光免疫细胞化学染色比较DNA-PKcs、FHIT、Id2、NFκB/p65及p16在SP细胞与非SP细胞中表达的差异。结果:胃癌SGC-7901细胞中SP细胞的比例为3.8%;FHIT、Id2及p16在SP细胞中的阳性表达率显著低于在非SP细胞中的阳性表达率(P<0.001),DNA-PKcs、NFκB/p65在SP细胞和非SP细胞中的阳性表达率无显著性差异。结论:FHIT、Id2及p16可能是胃癌SGC-7901癌干细胞的特异性分子标志物,而DNA-PKcs、NFκB/p65在鉴定胃癌干细胞时无特异性。
Objective: To explore specific molecular markers in side population of gastric carcinoma cell line SGC-7901. Methods: The percentage of SP cells was detected in gastric carcinoma cell line SGC-7901 by using Hoechst33342 staining, and the expression of DNA-PKcs, FHIT, Id2, NFKB/p65 and p16 in SP cells were compared with those in non-SP cells through fluorescence immunocytochemical method. Results: The percentage of SP cells in gastric carcinoma cell line SGC-7901 was 3.8%. The positive expression rates of FHIT, Id2 and p16 in SP cells were significantly lower than those in non-SP cells (P 〈0.001 ). There was no markedly difference in the positive expression rates of DNA-PKcs and NFKB/p65 between SP cells and non-SP cells. Conclusions: FHIT, Id2 and p16 may be specific molecular markers to cancer stem cells in gastric carcinoma cell line SGC-7901, while DNA-PKcs and NFKB/p65 may not relate to cancer stem cell.
