骨连蛋白对人晶状体上皮细胞在人工晶状体上黏附的影响 被引量:1

摘要 目的通过观察骨连蛋白的微量加入对晶状体上皮细胞黏附于人工晶状体表面的数目及形态特征的影响,探讨骨连蛋白有效抑制晶状体上皮细胞黏附的可行性。方法传代培养永生化的人晶状体上皮细胞系SRA01/04,分别制备含0mg·L-1、0.2mg·L-1、1.0mg·L-1、5.0mg·L-1的骨连蛋白完全培养液,然后滴加到嵌入琼脂板的人工晶状体上,在CO2培养箱中连续培养。分别于培养1d、3d、7d后,在倒置显微镜下观察细胞变化,并用血小板计数法计数人工晶状体表面黏附的细胞数,进行比较。结果浓度为0.2mg·L-1时骨连蛋白[作用1d、3d、7d后黏附的细胞数分别为(266.25±4.97)mm-2、(270.82±7.47)mm-2、(241.44±17.14)mm-2]有抑制黏附的趋势,但与0mg·L-1浓度组[作用1d、3d、7d后黏附的细胞数分别为(274.63±6.67)mm-2、(277.78±7·37)mm-2、(261.71±13.23)mm-2]相比差异无统计学意义(P>0.05);1.0mg·L-1骨连蛋白[作用1d、3d、7d后黏附的细胞数分别为(199.86±5.91)mm-2,(162.12±6.11)mm-2、(156.94±4.90)mm-2]即可见明确的抑制作用(P<0·01);5.0mg·L-1骨连蛋白[作用1d、3d、7d后黏附的细胞数分别为(180.30±7.37)mm-2,(141.16±10.83)mm-2,(130.41±13.50)mm-2]作用更加明显(P=0.00)。作用1d时各浓度组未见明确改变(P>0.05),3d、7d时可见明确的抑制作用(P<0·01)。结论骨连蛋白可以抑制晶状体上皮细胞在人工晶状体表面黏附,其作用浓度及作用时间呈现正相关性。 Objective To investigate the feasibility of osteonectin to inhibit human lens epithelial cell adhesion by observing the influence of microdose osteonectin on human lens epithelial cells number and morphologic character when they adhered to the artificial lens. Methods After serial subctfltivation of SRA01/04 ,0 mg·L^-1,0.2 mg·L^-1,1.0 mg·L^-1,5.0 mg·L^-1 osteonectin were added into cell suspension, respectively. Then the four samples were dropped to the artificial lens which inset put to CO2 incubaton for 1 day, 3 days, 7 into Agar. And the artificial lens were days. The change of lens epithelium cells were observed by inverted microscope and the cell population which attached to artificial lens were recorded. Results 0.2 mg·L^-1 osteonectin[ acting for 1 day, 3 days, 7 days, the numbers of adhered cells were ( 266.25 ± 4.97 ) mm^-2, ( 270. 82 ± 7.47 )mm ^-2, ( 241.44 ± 17.14 )mm ^-2, respectively ] had a tendency to inhibit the adhesion of lens epithelia, but it was no sense in statistics compared to the 0 mg·L^-1 group[ acting for 1 day,3 days,7 days,the numbers of adhered cells were (274.63 ± 6.67) mm^-2, (277.78 ± 7.37) mm^-2, (261.71 ± 13.23) mm^-2 ,respectively;P〉0. 05 ]. 1.0 mg·L^-1 osteonectin[ acting for 1 day,3 days,7 days,the numbers of adhered cells were( 199. 86 ± 5.91 )mm^-2, ( 162. 12 ± 6. 11 ) mm^-2, (156. 94 ± 4.90)mm^-2 ,respectively ] had a sure effect on inhibiting adhesion, and it was more significant when the concentration came to 5.0 mg·L^-1 acting for 1 day, 3 days, 7 days, the numbers of adhered cells were ( 180.30 ± 7. 37)mm^-2, (141.16 ±10. 83)mm^-2 , (130.41 ± 13.50) mm^-2 ,respectively;P 〈 0. 01]. The inhibiting effect were notable after culturing for 3 days and 7 days ( P 〈 0. 01 ). Conclusion Osteonectin can inhibit lens epithelium cells from adhering to artificial lens,and this effect showed direct correlation to the concentration and time.
