通络生骨胶囊对骨髓基质干细胞成骨和成脂肪双向分化的影响 被引量:12

摘要 目的:研究通络生骨胶囊对体外培养糖皮质激素诱导骨髓基质干细胞(MSCs)成骨和成脂肪双向分化的影响。方法:使用地塞米松浓度10-8M诱导MSCs成脂肪和成骨双向分化,以通络生骨胶囊治疗,采用逆转录聚合酶链反应法检测脂蛋白脂酶,骨桥蛋白基因的表达水平,观察MSCs成骨和成脂肪分化的改变。结果:地塞米松10-8M可诱导MSCs成骨和成脂肪双向分化;通络生骨胶囊具有明显的抑制成脂肪分化,促进成骨分化的作用。结论:通络生骨胶囊治疗激素性股骨头坏死药效机制,可能是通过对MSCs成脂肪和成骨分化调控的影响而产生作用。 Objective: To study the effect of Tongluoshenggu capsule on adipocytic and osteogenic differentiation in rat marrow stem cell. Methods: 10-8M dexamethasone was used to induce the bio-differentiation of MSCs and Tongluoshenggu capsule was taken in the treatment. The expression of lipoprotein lipase mRNA and osteopontin gene mRNA was determined by RT-PCR, the differentiation of MSCs osteogenic and adipocytic was observed. Results: The bio-differentiation of MSCs could be induced by 10-8M dexamethasone. The adipocytic process was inhibited by Tongluoshenggu capsule markedly and the osteogenic process was accelerated. Conclusion: The mechanism of Tongluoshenggu capsule on treating hormone-induced necrosis of femoral head might be its effect on MSCs adipocytic and osteogenic differentiation.
