摘要
[目的]弄清川西北高原野生垂穗披碱草与所建指纹库材料的遗传差别。[方法]对采集于川西北高原、新疆、西藏、青海的55份垂穗披碱草材料采用酸性聚丙烯酰胺凝胶电泳技术(A-PAGE),建立醇溶蛋白指纹库,将川西北高原材料的醇溶蛋白指纹与指纹库的进行对比分析。[结果]从川西北高原材料中分离出41条带纹,多态率为87.80%,少于所建指纹库材料(45条带纹,多态率93.33%)。Shannon指数、Simpson指数、遗传相似系数也小于所建指纹库材料,表明川西北高原材料的醇溶蛋白遗传多样性水平低于平均水平。川西北高原材料的特有带纹条数(2条)占指纹库共有带纹条数(5条)的40%,说明川西北高原垂穗披碱草具有自己特有的遗传特征。[结论]川西北高原材料可聚为一类,其醇溶蛋白图谱类型与材料所在的海拔有一定的相关性。
[ Objective] The aim was to ravel the genetic difference between wild Elymus nutans in the northwestern plateau of Sichuan Province and the materials from the established fingerprinting database. [ Method] Fifty-five shares of E. nutans materials collected from northwest plateau of Sichuan Province, Xinjiang, Tibetr and Qinghai were used to establish the gliadin fingerprinting database by A-PAGE technique. The gliadin fingerprints of the materials from northwest plateau of Sichuan Province were compared with that from the established fingerprinting database. [ Result ] Forty-one bands were separated from materials from northwest plateau of Sichuan Province and 87.80% of them were polymorphic, which was fewer than that from materials from the established fingerprinting database (45 bands and 93.33% of them were polymorphic). The Shannon index, Simpson index and genetic similarity coefficient were also less than that of materials from the established fingerprinting database, suggesting that the gliadin genetic polymorplfism level in the materials from northwest plateau of Sichnan Province was lower than the average level. The particular bands (2 bands) of materials from northwest plateau of Siehuan Province accounted for 40% of total bands (5 bands) in fingerprinting database, which showed that the E. nutans in northwest plateau of Sichuan Province had its particular genetic features. [Conclusion] The materials from northwest plateau of Sichuan Province could be clustered into a group and its gliadin map type had some correlation with the altitude at which the materials were.
