摘要
目的建立HPLC-ELSD测定青蒿素片中青蒿素含量的方法,并用于青蒿素片中青蒿素含量的测定。方法采用蒸发光散射检测器对青蒿素片中青蒿素进行HPLC分析,Thermo C18色谱柱(4.6mm×250mm,5μm);以甲醇-水(80:20)为流动相;流速为1.0mL.min-1;柱温为25℃;蒸发光散射检测器漂移管温度60℃,载气(N2)压力5.8×10-3Pa,增益值为50。结果青蒿素在1.002～4.008μg内呈现良好的线性关系(r=0.9996),平均加样回收率为99.45%(RSD为2.3%)。结论本法简单、准确、重复性好,可用于测定青蒿素片中青蒿素的含量。
OBJECTIVE To establish a HPLC-ELSD method for the determination of the content of artemisinin in artemisinin tablets.METHODS The analytical column was Thermo RP-C18（4.6 mm×250 mm,5 μm）.The mobile phase was methanol-water（80∶20）.The flow rate was 1.0 mL·min^-1.The samples were determined by an evaporative light-scattering detector（ELSD） Waters 2420 with the drift tube temperature at 60 ℃ and the nitrogen flow pressure of 5.8×10^-3 Pa.RESULTS The calibration curve was linear in the range of 1.002～4.008 μg（for artemisinin,r=0.999 9）.The average recovery（n=5） was 99.45%（RSD=2.3%）.CONCLUSION The method is simple,accurate and with good reproducibility.It can be used as the quality control of artemisinin tablet.
