摘要 探讨醛固酮(Ald)对血管平滑肌细胞蛋白合成、胶原合成及分泌的影响。方法将培养的乳鼠VSMC分成17组:正常对照组;Ald10-7组;安体舒通(Spi)10-7mol/L组;血管紧张素Ⅱ(AngⅡ)10-7mol/L组;碱性成纤维细胞生长因子(bFGF)10ng/L组;上述浓度的AngⅡ和bFGF分别加入不同浓度的Ald和Spi(10-9,10-8,10-7mol/L)组成不同浓度的配伍组。观察3H-亮氨酸和3H-脯氨酸参入的影响,以计算对蛋白质和胶原合成的情况。结果Ald明显增加VSMC的3H-脯氨酸参入,并加强AngⅡ及bFGF剌激的3H亮氨酸和或3H-脯氨酸的参入及胶原分泌。Spi抑制VSMC的3H-亮氨酸和或3H-脯氨酸的参入并抑制AngⅡ及bFGF的3H-亮氨酸和3H-脯氨酸的参入和胶原的分泌。结论Ald作为局部因子,通过旁分泌或自分泌参与VSMC的蛋白合成、胶原合成和分泌。 Aim\ To investigate the effect of aldosterone on the the protein,colagen in cultured rat vascular smooth muscle cells.\ Methods\ Protien synthesis was expressed by 3H leusine incorporation,the collagen by 3H proline in coporation in cultured cell.\ The cultured medium were added with the aldosterone(Ald) 10 7 mol/L alone,spironolactone(Spi) alone,angiotensinⅡ(AngⅡ) alone, basic fibroblast growing fortor(bFGF) alone, bFGF+Ald(in various concentrations), bFGF+Spi(in various concentrations).\ Results\ Adding of Ald increased the incorporation of 3H leucin, 3H proline in cultured vascular muscle cells; and increased the collagen secretion induced by simultaneously adding of AngⅡ and bFGF.\ Spi inhibited the incooporation and the secretion of collagen by adding AngⅡ and bFGF as well.\ Conclusion\ Ald involved in the protien synthesis,collagen synthesis and the collagen secretion. It may serve as a local important factor.
