摘要
目的:建立测定海洋抗肿瘤活性成分海参皂苷Nobiliside-A的HPLC含量测定方法。方法:通过比较紫外检测器和蒸发光散射检测器以及不同的色谱柱、不同的流动相系统,确定了Nobiliside-A的HPLC-ELSD含量测定方法,并对线性范围、精密度、稳定性进行了考察。结果:采用蒸发光散射检测器,Agilent Zorbax SB-C18色谱柱(150mm×4.6mm,5μm),流动相为甲醇:0.5%HAC(78:22),在0.273—4.24 μg范围内Nobiliside-A进样量的自然对数与峰面积的自然对数呈良好的线性关系(r=0.9995),日内、日间精密度以及稳定性均达到要求。结论:该实验为海参皂苷Nobillside-A的检测提供了准确、快速的分析方法。
AIM : To establish the assay method of Nobiliside-A, an anticancer active component in sea cucumber, by HPLC. METHODS: Comparative experiment was carried out between different detector and different chromatographic column and different mobile phase, at last a HPLC-ELSD method was determined, and linear range, precision and stability were studied. RESULTS: ELSD detector and Agilent Zorbax SB-C18 column were used, mobile phase was methonal: 0.5% HAC (78:22) , the natural logarithm of content of Nobiliside-A and the natural logarithm of the peak area were linear in the range of 0. 273 - 4.24 μg ( r = 0. 999 5 ), stability and precision within day and between day were satisfied. CONCLUSION: The experiment has provided a rapid and precise method for determining the content of Nobiliside-A.
