摘要
目的从鲍鱼菇(Pleurotus abalonus)子实体中分离纯化均一组分的多糖,初步研究该多糖对小鼠的抑瘤作用和免疫功能的影响。方法粗多糖经热水提取,95%乙醇沉淀,Sevag法去蛋白,DEAE-sephadex A-25柱层析纯化,获得均一组分的鲍鱼菇子实体多糖PPA-1。以瘤重大小判断抑瘤效果,以巨噬细胞吞噬功能检测非特异性免疫功能,以血清溶血素IgM的水平及迟发型超敏反应(DTH)检测特异性免疫功能。结果PPA-1≥75 mg/kg对小鼠S180移植性肿瘤有抑制作用(P<0.01);PPA-1≥50 mg/kg能提高荷瘤小鼠的脾指数(P<0.05、P<0.001),能提高正常小鼠巨噬细胞吞噬率(P<0.05、P<0.01)、吞噬指数(P<0.05、P<0.001)及血清溶血素IgM的水平(P<0.05);PPA-1≥75 mg/kg能增强正常小鼠迟发型超敏反应(P<0.05、P<0.001)。结论一定剂量的鲍鱼菇子实体多糖PPA-1能够抑制小鼠S180肿瘤生长,能够增强小鼠的非特异性免疫及特异性的体液免疫和细胞免疫功能。
Crude polysaccharides from the fruit body of Pleurotus abalonus were isolated by a series of sequential steps including extraction with hot water, precipitation with 95 % ethanol, removal of protein by Sevag method. Two water-soluble polysaceharides (PPA-1, PPA-2) were obtained after DEAE Sephadex A-25 anion-exchange chromatography. PPA-1 was of homogeneous by polyacrylamide gel electrophoresis and Sephadex G-100 gel filtration chromatography. The infrared spectrum analysis showed that PPA-1 had polysaccharides specific absorbent peak,and in the UV spectrum specific absorbent peak of nucleic acid and protein were not found. In vivo experiments showed that PPA-1 (75 mg/kg, 100 mg/kg) has antitumor activities on the implanted Sarcoma 180 in mice(P〈0.01), PPA-1 (50 mg/kg, 75 mg/kg, 100 mg/kg)can increase the spleen index in tumor-bearing mice(P〈0.05, P〈0.001)and enhance the phagocytic rate(P〈0.05, P〈0.01)and the phagocytic index(P〈 0. 05, P〈0. 001)of celiac macrophage, production of serum hemolysins IgM (P〈0.05) in mice ,PPA-1 (75 mg/kg,100 rag/ kg) can enhance the function of delay hypersensitivity induced by DNFB(P〈0.05,P〈0.001) in mice . It is concluded that a certain dosage of PPA-1 could inhibit the transplanted tumor growth of S180 ,improve non- specific immune function and specific immune function such as humoral immunity, cellular immunity in mice.
