摘要
目的:探讨白藜芦醇及其联合阿糖胞苷对K562细胞凋亡的影响及相关机制。方法:采用免疫组织化学法和RT-PCR法分别检测白藜芦醇及其联合阿糖胞苷处理K562细胞48h后Fas、FasL和Survivin表达情况。结果:K562细胞分别经白藜芦醇、阿糖胞苷处理48h后白藜芦醇组和阿糖胞苷组Fas表达光密度值较对照组升高(P<0.05),FasL表达上调较对照组有显著性变化(P<0.05),两药联合作用Fas表达较单用白藜芦醇无显著性变化(P>0.05),而FasL表达上调较单用白藜芦醇或阿糖胞苷有显著性变化(P<0.05)。白藜芦醇、阿糖胞苷单独及联合作用48h后,K562细胞的Survivin mRNA表达较细胞对照组下降(P<0.05、P<0.01).两药联合作用Survivin mRNA表达较单用白藜芦醇无显著性变化(P>0.05)。结论:白藜芦醇单独及联合阿糖胞苷能通过上调Fas/FasL的表达,激活Fas/FasL凋亡途径,诱导人白血病K562细胞凋亡,同时下调Survivin的表达促进凋亡的发生。
Objective:This study was aimed to explore the apoptotic effect the K562 cells treated by resveratrol and/or arabinosylcytosin. Methods:Fas/FasL expression was detected by immunohistochemical method and survivin expression was detected by RT-PCR in K562 cells, 48 hours after treatment by resveratrol and arabinosylcytosin respectively. Results: Fas/FasL expression in K562 cells,treated by resveratrol and arabinosylcytosin respectively, was significantly increased compared with the control group(P〈0.05). However, the effect of resveratrol combined with arabinosylcytosin in K562 cell Fas expression showed no significance changes in comparison with resveratrol or arabinosylcytosin(P〉0.05),contrary to FasL expression (P〈0.05). Survivin mRNA expression was decreased after 48 hours in K562 cells treated by resveratrol and/or arabinosylcytosin in comparison with the control group(P〈0.05,P〈0.01 ). However, the effect of resveratrol combined with arabinosylcytosin in K562 cell Survivin mRNA expression showed no significance changes in comparison with resveratrol or arabinosylcytosin(P〉0.05 ). Conclusion:In conclusion the resveratrol and/or arabinosylcytosin could upregulate Fas/FasL expression and activate the apoptosis pathway of Fas/FasL to induce the apoptosis of K562 cells. It could downregulate the expression of survivin to promote apoptosis. The two drugs had synergistic effect.
