摘要
目的从基因水平探讨载脂蛋白E(ApoE)基因多态性与中国北方汉族人血脂的关系。方法应用聚合酶链反应-限制性片段长度多态性(PCR-RFLP)技术测定150例心血管病患者(病例组)和130例正常对照者(对照组)的ApoE基因多态性,并测定血脂代谢的指标,研究ApoE基因多态性与血脂水平的关系。结果在3种等位基因间,总胆固醇(TC)、低密度脂蛋白胆固醇(LDL-C)及载脂蛋白B(ApoB)水平均按ε4基因携带者>ε3基因携带者>ε2基因携带者的顺序递减,差异有显著性(对照组均P<0.05,病例组均P<0.01),而三酰甘油(TG)、高密度脂蛋白胆固醇(HDL-C)、载脂蛋白AI(ApoAI)水平差异无显著性(均P>0.05)。结论ApoE基因多态性影响中国北方汉族人的血脂代谢,ε4等位基因对TC、LDL-C及ApoB有升高作用。
[Objective] To study the relationship between polymorphisms of apolipoprotein E(ApoE) gene and serum lipids in Northern Chinese Han population by gene level.[Methods] ApoE gene polymorphism was genotyped in 150 cardiovascular patients (patient group) and 130 healthy controls (control group) by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) method. Meantime, the levels of serum lipids were measured. [Results]In comparison of the levels of serum lipids among three kinds of Apo E allele, the levels of TC, LDL-C and ApoB were decreased progressively according to ε4 allele 〉ε3 allele 〉ε2 allele, which had a statistically difference (all P 〈0.05 in control group, all P 〈0.01 in patient group), while the levels of TG, HDL-C and ApoAI had no significant differentce among the three groups (all P 〉0.05). [Conclusions] ApoE gene polymorphism affects serum lipid metabolism in Northern Chinese Han population, and ε4 allele has a function of increasing TC,LDL-C and ApoB.
