摘要
目的:在TOP10大肠埃希菌pTrcHisB系统中表达人4-1BBL(h4-1BBL)融合蛋白,并对其进行鉴定。方法:应用RT-PCR方法,从Raji细胞中克隆h4-1BBL基因中的CDS序列,构建pTrcHisB-h4-1BBL原核表达载体,测序后,经IPTG诱导表达,Westernblot鉴定h4-1BBL融合蛋白的表达情况。结果:成功构建了重组载体pTrcHisB-h4-1BBL,重组蛋白是带6个His标记的h4-1BBL融合蛋白。结论:重组载体pTrcHisB-h4-1BBL成功表达了h4-1BBL融合蛋白,为进一步研究其功能奠定了一定的基础。
Objective:To express and identify the human 4-1BB ligand fusion protein in E. coll. Methods:RT-PCR was used to clone CDS sequence of human 4-1BB ligand gene from Raji cells and construct pTrcHisB-h4-1BBL prokaryotic expression vector. After sequencing ,the human 4-1BB ligand fusion protein was identified by Western blot assay. Results:Recombinant plasmid pTreHisB-h4- 1BBL was successfully constructed. The 6His-labeled human 4-1BB ligand fusion protein was expressed under induction of IPTG. Conclusions:Human 4-1BB ligand fusion protein was successfully expressed in pTrcHisB-h4-1BBL recombinant plasmid and it lays the foundation for further research on human 4-1BB ligand.
