摘要
骨桥蛋白(osteopontin,OPN)是一种分泌型磷酸化糖蛋白,其分子结构中含有精氨酸-甘氨酸-天冬氨酸(RGD)的受体结合序列。OPN主要通过与其受体整合素和CD44相互作用,参与肿瘤转移的多个过程。近年来,骨桥蛋白及其调控肿瘤侵袭与转移的信号通路倍受关注。OPN通过与整合素ανβ3和CD44结合作用,使多种激酶(PI3K、NIK、PLC、PKC和MAPK)磷酸化激活,从而发挥抑制凋亡和激活多种基质降解蛋白酶(uPA和MMPs)的功能,促进肿瘤细胞转移。
