摘要 目的探讨p73基因在鼻咽癌及癌旁组织中的表达与突变。方法采用定量逆转录聚合酶链反应(RT-PCR)技术检测35例鼻咽癌与癌旁组织中p73基因的表达;采用单链构象多态性技术(PCR-SSCP)检测鼻咽癌与癌旁组织中p73基因的突变情况。结果(1)定量RT-PCR检出p73基因在26例鼻咽癌组织中呈高表达,在2例癌旁组织中呈高表达。p73基因在鼻咽癌与癌旁组织中的表达率之间的差异具有显著性意义(P<0.01)。(2)等位基因表达分析示p73基因在14例杂合性鼻咽癌标本中检出10例存在G/C:A/T双等位基因表达,在癌旁组织中皆为G/C表达,未见A/T表达。(3)PCR-SSCP未检出p73基因突变。结论p73基因mRNA的表达可能与鼻咽癌的发生有关。 Objective To investigate the expression and mutation of p73 gene in nasopharyngeal carcinoma tissue and tumor-adjacent tissues. Methods The expression of p73 gene in 35 specimens of nasopharyngeal carcinoma and matched tumor-adjacent tissues were detected by quantitative RT-PCR, and the alteration of p73 gene was analyzed by SSCP. Results (1) Nasopharyngeal carcinoma tissues exhibited overexpression of p73 gene in 26 specimens. Matched tumor-adjacent tissues exhibited overexpression of p73 in 2 specimens. The expression rates of p73 gene were significantly different between nasopharyngeal carcinoma and tumor-adjacent tissues (P〈0.01). (2) Allele-specific analysis showed that expression of G/C: A/T existed in 10 of 14 heterozygous nasopharyngeal specimens, and expression of G/C existed in tumor-adjacent specimens. (3) No mutation in nasopharyngeal carcinoma and tumor-adjacent tissues was detected by SSCP. Conclusion The overexpression of p73 gene may be related to the occurrence of nasopharyngeal carcinoma.
