摘要
目的研究重组牛碱性成纤维细胞生长因子(bFGF)对糖尿病大鼠皮肤溃疡促愈合的机制。方法采用雄性wistar大鼠,制备糖尿病模型,造成背部皮肤缺损,将bFGF溶液连续15天喷注于创面,透明膜描记记录伤后第5、10、152、0天创面面积。伤后第51、0、15、20天取创面组织,观察其病理变化,包括肉芽组织的生长和再上皮化情况。结果bFGF可以明显加速糖尿病大鼠皮肤溃疡的愈合速度,使创面面积显著减小(P<0.05).组织学检查:bFGF治疗组在创伤10天成纤维细胞生长活跃,数量明显增多,成纤维细胞与毛细血管肉芽数量明显多于对照组;创伤后15天,创面收缩与再上皮化明显,新生上皮向创面中心爬行较快。在bFGF作用组,MMP-9和Timp-1表达增强。结论bFGF对于糖尿病大鼠皮肤溃疡的愈合有明显的促进作用。bFGF可能通过促进皮肤细胞高表达MMP-9和Timp-1途径加快皮肤损伤的修复。
Objective To investigate the effect of recombinant bovine basic fibroblast growth factor (rb-bFGF) in treating on dermal chronic ulcers of diabetic rats. Methods We prepared dermal chronic ulcers in diabetic rats. Randomly divided into 2 groups according to different treatment. The wounds of the experimental group were treated with rb-bFGF and the wounds of the control group were treated with physiological saline. The therapeutic results were assessed with healing area, healing rate, healing time and histomor- phological analysis were compared between the two groups. Results The healing rate in bFGF group were obviously higher than that in the control group ( P 〈 0.05). In the group treated with bFGF, healing area at different time points were significantly decreased compared with the control group (P 〈 0.05).bFGF resulted in the remarkable enhancement of granula6on tissue formation. Expression of MMP-9 and Timp- 1 increased in bFGF treated group. Conclusion Topical application of bFGF can significantly accelerate the healing of dermal chronic ulcers in diabetic rats and its mechanism maybe through overexpressed MMP-9 and Timp-1 in the skin.
