摘要
研究聚丙烯酰胺凝胶电泳(polyacrylamide gel electrophoresis,简称PAGE)鉴定脱墨浆(DIP)生产系统中过氧化氢酶的实验条件,并对各工段过氧化氢酶同工酶进行分析。实验结果表明,碎浆工段点样量3μL,一段浓缩点样量5μL,二段浓缩点样量10μL,浓缩胶质量分数4%,分离胶质量分数9%,才能得到较高质量的电泳结果。生产系统中共检测出6条酶带,各工段出现的酶带的数量不同,种类也不同,仅有一条酶带在三个工段均出现,探讨了此现象产生的原因并对6种同工酶分子量进行排序。
The experimental conditions of polyacrylamide gel electrophoresis (PAGE) to identify catalase were studied and isoezyme of catalases in each stage were analyzed in deinked pulp (DIP) production systems. The results indicate that the linear range of repulping stage is 3μL, the effluent liquor after the first thickening is 5 μL and the effluent liquor after the second thickening was 10 μL, the isolation gel density was 9% and the concentration gel density was 4%. According to the results, there are 6 enzyme bands identified in production system, with each section having different enzyme bands in quantities and species, only one bend appeared in all the three stages. The reasons arising from phenomenon above was researched and the order of molecular size of isoezyme was done.
