摘要
[目的]为籼粳稻亚种的分子标记鉴定提供依据。[方法]以13份籼稻、5份粳稻为亲本,采取SDS法提取水稻叶片总DNA,进行扩增反应,利用321对SSR引物对亲本的多态性进行筛选,分析籼粳亚种之间的SSR标记差异。[结果]18个亲本中共筛选到168对引物具有明显的标记带和较好的多态性。SAR水稻血缘的412、407、421、992、950之间及长药野生稻血缘的84-15与84-23之间的多态性明显少于稳定亲本与栽培稻之间的多态性,也低于籼稻品种间和粳稻品种间的多态性。籼稻与粳稻之间的多态性频率最高,籼稻品种之间的多态性高于粳稻品种之间的多态性。供试材料籼粳稻之间存在31个SSR标记差异,SSR标记可较为准确地鉴定材料的亚种属性。[结论]籼粳亚种之间有明显的SSR标记差异。
[Objective] The study aimed to supply basis for the molecular marker identification ofindica-japonica rice subspecies. [Method] With 13 shares of indica rice and 5 shares of japonica rice as parents, the total DNA in rice leaf was extracted by SDS method for amplification reaction and the polymorphism of parents was screened by 321 pairs of SSR primers. The SSR marker difference among indica-japonica subspecies was analyzed. [Result] 168 pairs of primers with obvious marker-bands and better polymorphism were screened out from 18 parents totally. The polymorphisms among 412, 407, 421,992 and 950 derived from rice SAR and between 84-15 and 84-23 derived from Oryza longistaminata were obviously less than that between stable parents and cultivated rice and also lower than that among indica rice varieties and japonica rice varieties. The polymorphism frequency between indica rice and japonica rice was highest and that among indica rice varieties was higher than that among japonica rice varieties. There were 31 SSR marker differences between indica rice and japonica rice of tested materials and SSR marker could identify subspecies attribute of material more accurately. [Conclusion[ There was obvious SSR marker difference among indica-japonica subspecies.
