摘要 目的:建立柱后衍生-高效液相色谱法测定保健食品、强化食品和营养素补充剂中的叶酸。方法:样品经水提取,选用乙腈+50 mmol/L磷酸二氢钾溶液(pH=3.5)=12+88为流动相,流速为1.0 ml/min,经Kromasil C18柱分离(柱温为常温),用0.5%的过二硫酸钾为衍生剂,流速0.3 ml/min,反应温度60℃,使用荧光检测器,Ex=365 nm,Em=450 nm。结果:被测组分浓度(0.010~2.0μg/ml)与峰面积线形关系良好(r=0.9999);加标回收率为92.11%~95.31%,相对标准偏差(RSD)2.99%~3.44%。结论:该方法操作简便、快速、测定结果准确可靠,适合保健食品、强化食品和营养补充剂中的叶酸测定。 Objective:A quantitative method was developed for the analysis of folic acid in health foods, supplementary foods by high performance liquid chromatographic method with post - column chemical derivation and fluorescence detection. Methods: After being extracted with water in an uhrasonica,a Cls column was used for separation at room temperature,and elution was performed with acetonitrile + potassium dihydrogen phosphate( pH = 3.5) = 12 + 88 at 1.0 ml/min. Folic acid in the eluate was deri- vatized to an intensive fluorescence compound by 0, 5% potassium peroxydisulfate at 60℃ and detected by a spectrofluorimetor (Ex = 365 nm, Em = 450 nm) at 0. 3 ml/min. Results: A linear calibration curve was obtained in the range of 0. 010 -2. 0μg/ml ( r = 0. 9999) ,RSD were 2. 99% - 3.44% and the recoveries of spiked analyte were in the range of 92. 11% - 95. 31%. Conclusion:The developed method can be applied to determine the folic acid contents in health food and supplementary foods. The method is rapid, simple, accurate and reliable.
