摘要
以纯化的谷胱甘肽转移酶标签蛋白(GST)免疫BALB/c小鼠,按常规方法进行细胞融合,用纯化的GST筛选,最终获得一株抗GST的杂交瘤细胞系(命名为1D10),染色体平均记数为90对,间接ELISA检测1D10细胞培养上清的效价为1:4 000。腹水效价为1:5×107,该单抗可以特异识别GST蛋白和带有GST标签(GST-tagged)的融合蛋白。
The monoclonal antibody (McAb) against GST was performed by immunizing the BABL/c mice with purified GST. Cells fusion was carried out by standard method and hybridmas secreting antibody were subcloned three times with limiting dilution. One hybridoma cell line named 1D10 was picked out by indirect ELISA, which secreted monoclonal antibody specific against GST, and its average number of chromosomes was 90 paries. The antibody titer of cell culture supernatant and ascite were 1:4000 and 1:5×10^7, respectively.
