摘要 利用海藻酸钙凝胶颗粒固定葡萄糖氧化酶(GOD)。确定了固定化条件,并考察了温度、pH值、储存时间对固定化酶和游离酶酶活力的影响,测定了酶活回收率。确定的固定化较优条件为:CaCl25.0%(质量体积比),海藻酸钠3.0%(质量体积比);固定化酶的最适催化温度为31℃、最适pH值为6.3,较游离酶分别提高7℃和0.6;固定化酶的平均酶活回收率为61.69%。此外,酶的储存稳定性能也有所提高,可重复多次使用。 In this study, calcium alginate gel capsule was chosen as the carrier to immobilize glucose oxidase (GOD). The optimal conditions for GOD encapsulation were obtained as CaCl2 5.0% (ratio of mass to volume) and sodium alginate 3.0% (ratio of mass to volume), and the effects of temperature, pH value, storage time on the activity of free and immobilized GOD were studied. Furthermore, the recovery rate of immobilized GOD activity was investigated. The results showed that the optimum pH value and temperature for immobilization GOD were 6.3 and 31℃, respectively,which enhanced 0.6 and 7℃ compared to that of free enzyme. And the average recovery rate of immobilized GOD activity was 61.69 %. Moreover, immobilized enzyme had good storage stability and could be used repetitiously.
