几丁质固定化菊粉酶的研究 被引量:7

摘要 以自制的几丁质作载体,戊二醛作交联剂,用吸附交联法对菊粉酶进行了固定化研究。优化了固定化反应条件,在最适条件下,菊粉酶活力最大收率为32%。固定化菊粉酶的最适温度为60~65℃,比游离酶提高5℃。最适pH保持不变。固定化酶的热稳定性和对酸碱稳定性都有较明显的改善,将固定化酶装柱进行菊芋汁连续水解试验,操作半衰期达22d。 Inulinase was immobilized on Chitin,with glutaraldeyde as crosslinking agent. Theconditions of immobilization were sdudied systematically,and under the optimal conditions,the enzyme activity recorery of the immobilized inulinase was 33%. The optimal temperatureof the immobilized enzyme was at the range of 60~ 65℃ which was higher than that of thesoluble inulinase by 5℃. The optimal pH was not changed by immobilization. Thermal stabil-ity of the inulinase was improved by immobilizing treatment. so did the pH stability. Underthe condition of 6O℃,the half-life of the immobilized inulinase was 22 days.
