L-精氨酸对兔肺缺血再灌注时血红素氧合酶-1的影响 被引量:1

Effect of L-arginine on heme oxygenase-1 in the lung after pulmonary ischemia-reperfusion in rabbits

摘要 目的探讨L1广精氨酸对兔肺缺血再灌注时血红素氧合酶-1(HO-1)的影响。方法健康日本大耳白兔20只,雌雄不拘,体重1.7~2.6kg,随机分为2组,肺缺血再灌注组(IR组)和L-广精氨酸治疗组(L-Arg组),每组10只。建立兔单肺原位缺血再灌注损伤模型,L-Arg组肺缺血前20min耳缘静脉注射LArg溶液100mg/kg。采用原位杂交方法测定兔肺组织HO-1的表达;计算肺组织湿/干重比(W/D)及损伤肺泡百分数(IAR);电镜下观察肺组织超微结构。结果HO-1在两组肺血管内皮、部分血管平滑肌、外膜层及部分气道上皮均有表达;与IR组比较,L-Arg组HO-1水平升高,W/D及IAR降低(P〈0.05);L-Arg组肺组织形态学异常改变轻于IR组。结论L广精氨酸可通过上调肺组织HO-1的表达,增强肺组织HO-1的活性,减轻兔肺缺血再灌注损伤。 Objective To investigate the effect of L-arginine (L-Arg) on the expression and activity of heine oxygenase-1 (HO-1) in the lung tissue after pulmonary ischemia-reperfusion (I/R). Methods Twenty healthy Japanese long-ear white rabbits of both sexes weighing 1.7-2.6 kg were randomly divided into 2 groups (n = 10 each): group Ⅰ pulmonary I/R and groupⅡ L-Arg + I/R. The animals were anesthetized, tracheostomized and mechanically ventilated. Lung ischemia was induced by ligation of the blood vessels and bronchus in the hilum of the left lung for 1 h followed by 3 h reperfusion according to Sekido. In L-Arg group the animals received L-Arg 100 mg/kg 20 rain before ischemia. The animals were killed at the end of 3 h reperfusion and the lungs were removed for determination of HO-1 expression in the lung, W/D lung weight ratio and the injured alveoli rate (IAR) and microscopic examination with light and electron microscope. Results HO-1 expression was detected in the pulmonary endothelial cells, pulmonary vascular smooth muscle, extima of vessels and epithelial cells of airway in both groups. The absorbance and activity of HO-1 were significantly higher while W/D ratio and IAR values were significantly lower in L-Arg group than in I/R group. The morphological abnormality was less severe in L-Arg group than in I/R group.Conclusion L-Arg can protect the lung against I/R injury by up-regulating HO-1 expression and increasing HO-1 activity in the lung.
