摘要
以乙醇/二乙胺(100/0.1)为流动相,用纤维素三苯基氨基甲酸酯涂敷型手性固定相在模拟移动床色谱(SMBC)上拆分了奥美拉唑对映体。考察了在保持柱子切换时间不变的情况下,流动相内部流速对分离效果的影响。实验条件下,通过有序地优化SMB各区的流量.S-奥美拉唑的纯度、回收率均大幅度提高。使分离后S-奥美拉唑的纯度最高可达98.71%,R-奥美拉唑的纯度最高为95.47%。
The enantiomers of omeprazole were resolved by simulated moving chromatography on the coated cellulose triphenylcarbamate chiral stationary phase using ethanol and diethylamine (100/0.1) as the mobile phase. The effects of recycling flow rate on the separation were investigated when the columns switching time were kept constant. Under the experimental conditions, by optimizing the flows of SMB, the concentration, productivity of (s)-omeptazole were increased, we gain the satisfactory results which the highest purity of (s)-omeptazole is 98.71%,and the highest purity of (r)-omeptazole is 95.47%.
